ASO RNA and siRNA are a powerful new class of therapeutic modalities, which can target previously untreatable disease states. Robust and easy to implement analytical methods have proved difficult to develop. This has been made more challenging by the initiative to remove HFIP, a common reagent in the eluent system, from use in new methodologies. Most approaches for this remain inadequate for comprehensive characterization.  Here we describe a GLP compliant answer to this problem that has proven up to the task and extremely versatile in the ability to characterize a range of different oligonucleotides. Many new drug candidates can appear in manufacturer pipelines and in clinical trials. To ensure high quality, safety, and efficacy, analytical methods need to be developed quickly and be easy to perform.  The breakthroughs described here will aid in getting these new medicines to the market.
