SureQuant Hybrid DIA - Instrument method setup, data processing, result review

Combining a targeted peptide quantitative analysis with a data-independent acquisition analysis is gaining popularity. Such approaches are referred to as “hybrid DIA” methods (e.g., targeted MS2 hybrid DIA, SureQuant hybrid DIA). They offer sensitive and accurate quantification of selected peptides (based on targeted scans) while simultaneously allowing for a proteome-wide discovery (based on DIA scans).

This free online workshop will introduce the SureQuant method, a strategy for accurate quantification of endogenous peptides in an experiment where the targeted scan is triggered following the detection of isotopically labeled peptide standards added to the sample before the analysis. We will review details of instrument method setup on Thermo Scientific Orbitrap Astral mass spectrometer and walk step-by-step through the data processing steps in Proteome Discoverer.

The focus of the workshop is on workflows and parameters related to targeted peptide quantification. The participants will be able to request example .RAW file to complement the material presented during the workshop (requires PD 3.3 version).

The workshop will be recorded and made available to all registered participants. The presentation slides, featuring a step-by-step guide to processing SureQuant hybrid DIA data files, can be downloaded after the session.

The workshop is aimed at both novice and intermediate users. We have planned in time to answer your questions.
